Thymosin Alpha-1 (Tα1) is a 28-amino acid acetylated peptide originally isolated from thymosin fraction 5 by Allan Goldstein at George Washington University in 1977. It is naturally produced by thymic epithelial cells and functions as a master regulator of adaptive immunity. Tα1 acts primarily through Toll-like receptors TLR2 and TLR9 on dendritic cells, driving their maturation and enhancing antigen presentation to naïve T-lymphocytes. This cascade promotes T-cell differentiation into Th1-polarized effector cells, which are critical for antiviral and antitumor immunity.

1. Tα1 binds TLR2 and TLR9 on plasmacytoid dendritic cells, triggering MyD88-dependent signaling and NF-κB activation.
