Cardiolipin Targeting: A Precision Mitochondrial Approach

SS-31 (Elamipretide, D-Arg-Dmt-Lys-Phe-NH&sub2;) is a cell-permeable tetrapeptide that selectively accumulates in the inner mitochondrial membrane through electrostatic interaction with cardiolipin, a unique diphosphatidylglycerol lipid found exclusively in this compartment. Cardiolipin is essential for cristae structure, electron transport chain supercomplex assembly, and cytochrome c anchoring. SS-31 stabilizes cardiolipin’s interaction with respiratory complexes, restoring electron transfer efficiency without acting as a classical uncoupler.

1. SS-31 concentrates approximately 1,000-fold in mitochondria relative to cytoplasm within 30 minutes of exposure, driven by the organelle’s negative membrane potential and the peptide’s cationic charge.
2. Once localized, it binds cardiolipin at the inner membrane and prevents peroxidation of the unsaturated acyl chains that are critical for membrane curvature and cristae folding.
