This discovery (Lee et al., Cell Metabolism, 2015) overturned the longstanding assumption that mitochondrial DNA only encodes for 13 electron transport chain subunits, 22 tRNAs, and 2 rRNAs. MOTS-c is encoded within the 12S rRNA gene and is translated in the cytoplasm after mitochondrial mRNA export. Its existence reveals a previously unknown mitochondria-to-nucleus communication pathway that regulates metabolic homeostasis.

  1. MOTS-c is translated from an open reading frame within the mitochondrial 12S rRNA gene (MT-RNR1) on a cytoplasmic ribosome.
  2. Under metabolic stress, MOTS-c translocates to the nucleus where it interacts with AMPK and regulates the expression of antioxidant response element (ARE)-driven genes.
  3. In skeletal muscle, MOTS-c activates AMPK, increasing glucose uptake via GLUT4 translocation independently of insulin signaling.
  4. MOTS-c inhibits the folate-methionine cycle by suppressing AICAR transformylase, redirecting one-carbon metabolism and activating AMPK through AICAR accumulation.
  5. Circulating MOTS-c levels are exercise-responsive (increasing 50–100% after acute exercise) and decline with aging in both mice and humans.
