First-Generation Growth Hormone Secretagogue

GHRP-6 (Growth Hormone Releasing Peptide-6, His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) was the first synthetic GH secretagogue developed, pioneered by Cyril Bowers in the 1980s. It activates the growth hormone secretagogue receptor 1a (GHS-R1a)—now known as the ghrelin receptor—which was subsequently discovered to be the endogenous receptor for the “hunger hormone” ghrelin. GHRP-6 was instrumental in the discovery of ghrelin itself: the receptor was identified through GHRP-6 binding studies before the natural ligand was found.
