Second-Generation Growth Hormone Secretagogue

GHRP-2 (Growth Hormone Releasing Peptide-2, D-Ala-D-2Nal-Ala-Trp-D-Phe-Lys-NH2) is a second-generation hexapeptide GH secretagogue with the highest GH-releasing potency of any peptide in its class. Compared to GHRP-6, GHRP-2 produces approximately 30–50% greater GH release per microgram while generating significantly less appetite stimulation and ghrelin-like hunger signaling. This improved selectivity profile made GHRP-2 the preferred research GH secretagogue when appetite stimulation is undesirable.

1. GHRP-2 binds GHS-R1a with higher affinity than GHRP-6, producing more potent and consistent GH release from pituitary somatotrophs.
2. The D-2-naphthylalanine at position 2 provides enhanced receptor binding stability and reduced cross-reactivity at appetite-stimulating hypothalamic sites.
3. GH pulse amplitude from GHRP-2 is the highest of any GHRP: peak levels of 30–60 ng/mL are routinely achieved at standard research doses.
