Its mechanism involves broad neuroendocrine regulation rather than direct sedation.

  1. DSIP modulates GABA-A receptor subunit expression in the hypothalamic preoptic area, enhancing the sleep-promoting ventrolateral preoptic nucleus (VLPO) output.
  2. Cortisol and ACTH secretion rhythms are normalized through direct hypothalamic action, restoring the cortisol nadir during early sleep that is disrupted in insomnia and stress disorders.
  3. Somatotroph axis signaling is enhanced: DSIP increases nocturnal growth hormone pulse amplitude, consistent with enhanced slow-wave sleep (the GH secretory window).
